Seryl-tRNA Synthetases: Enzymes with Multiple Personalities*
نویسندگان
چکیده
Seryl-tRNA synthetases (SerRS) are essential enzymes responsible for aminoacylation of cognate transfer-RNAs (tRNAs) with serine, which is then incorporated into the proteins being synthesized on the ribosome. Although tRNA synthetases are usually a very conserved class of enzymes, phylogenic analyses revealed the existence of two distinct types of serine-charging enzymes; a standard or bacterialtype SerRS is found in the majority of organisms (prokaryotes, eukaryotes and archaea), while a highly diverged methanogenic-type SerRS is confined to the methanogenic archaea. We give a short overview on our recent biochemical and structural contributions to the understanding of different molecular mechanisms of tRNA serylation.

Trypanosoma seryl-tRNA synthetase is a metazoan-like enzyme with high affinity for tRNASec.
Trypanosomatids are important human pathogens that form a basal branch of eukaryotes. Their evolutionary history is still unclear as are many aspects of their molecular biology. Here we characterize essential components required for the incorporation of serine and selenocysteine into the proteome of Trypanosoma. First, the biological function of a putative Trypanosoma seryl-tRNA synthetase was ...

Two temperature-sensitive mutants of Escherichia coli have been found in which the conditional growth is a result of a thermosensitive leucyl-transfer ribonucleic acid (tRNA) synthetase and seryl-tRNA synthetase, respectively. The corresponding genetic loci, leuS and serS, cotransduce with lip and serC, respectively. As a result of the mutationally altered leucyl-tRNA synthetase, some leucine-,...

Class 2 aminoacyl-tRNA synthetases, which include the enzymes for alanine, aspartic acid, asparagine, glycine, histidine, lysine, phenylalanine, proline, serine and threonine, are characterised by three distinct sequence motifs 1,2 and 3 (reference 1). The structural and evolutionary relatedness of these ten enzymes are examined using alignments of primary sequences from prokaryotic and eukaryo...
متن کاملA specific hepatic transfer RNA for phosphoserine.
Radioactive O-phosphoryl-L-serine was detected after alkaline deacylation of rat and rooster liver [(3)H]seryl-tRNA acylated in vitro with homologous synthetases. Ribonuclease treatment of this tRNA yielded a compound with the properties of phosphoseryl-adenosine. Benzoylated DEAE-cellulose chromatography of seryl-tRNA yielded four distinct peaks, only one of which contained phosphoserine. A un...
متن کاملSeryl-tRNA Synthetases in Translation and Beyond
For a long time seryl-tRNA synthetases (SerRSs) stood as an archetypal, canonical aminoacyl-tRNA synthetases (aaRS), exhibiting only basic tRNA aminoacylation activity and with no moonlighting functions beyond protein biosynthesis. The picture has changed substantially in recent years after the discovery that SerRSs play an important role in antibiotic production and resistance and act as a reg...
